Cover lletter
Sometimes, the publisher/agent will ask for specific information about yourself but for the most part, describe the story/book you are submitting.
This site offers some tips...but I'm not for all the hyperbole
Elevator Pitch
If you are submitting a novel/novella then you should have one.

IMHO, that should be at the top of the letter. You have to get their attention and that is a key element in making your story stand out from the rest.
