The notion of a third gender has existed for centuries in different cultures worldwide. In some Native American cultures, the term “two-spirit” is an umbrella term describing those who fulfill a third gender.

Examining old writings
I read this wrong the first time. Sorry. This is from this side of the pond. Similar things happened in the Middle East, and India.
If you look at matthew 19:12 "For there are eunuchs who were born thus from their mother’s womb, and there are eunuchs who were made eunuchs by men, and there are eunuchs who have made themselves eunuchs for the kingdom of heaven’s sake. He who is able to accept it, let him accept it.”
Isaiah is particularly telling. Isaiah 56:4-5 4 For thus saith the Lord unto the eunuchs that keep my sabbaths, and choose the things that please me, and take hold of my covenant;
5 Even unto them will I give in mine house and within my walls a place and a name better than of sons and of daughters: I will give them an everlasting name, that shall not be cut off.
People become Transgender for many reasons. As long as one is honest and moral, there should be no issue. Transgender folk should value this.

Also I had never heard the term "muxe" before. Had you?

Trans-cultural Trans-gender
I am very interested in gender in other cultures. I have written about it is indigenous America ("Indian Red" and "Lumberjackie"), in India ("The Power of the Goddess", "Devotee" and maybe "Stiff Upper Lips") and also in Samoan culture ("Masina").
There is also evidence of it in ancient times as Gwen referred to - biblical references included. In the worship of female deities male priests would often castrate themselves to become female and be more true to their faith.
I have tried to build stories around some of these, but there is always room for more.
